Orphaned sisters Vi (Hailee Steinfeld) and Powder (Mia Sinclair Jenness) are taken in by Vander (JB Blanc), former rebellion leader in Zaun. Years later, Vi, Powder, and their adopted brothers rob a Piltover penthouse, but Powder accidentally triggers an explosion with stolen magical crystals, escalating tensions. Vander, now a community leader, tries to protect them from enforcers while crime lord Silco (Jason Spisak), whom Vander once betrayed, schemes using a powerful mutagen called Shimmer.
Jayce (Kevin Alejandro), a Piltover student, faces backlash for experimenting with arcane technology but finds an ally in Viktor (Harry Lloyd), Professor Heimerdinger’s (Mick Wingert) assistant. Vi tries to take responsibility for the robbery, but Vander is captured by Silco, whose mutated enforcer Deckard kills enforcer leader Grayson (Shohreh Aghdashloo). Vi, Mylo, and Claggor attempt a rescue, but Powder’s misguided help causes an explosion that kills Mylo and Claggor. Vander saves Vi but dies in the process. In grief, Vi lashes out at Powder, calling her a "jinx," leading Powder to embrace Silco as a father figure.
Years later, Piltover thrives with Jayce’s Hextech advancements. Powder, now Jinx (Ella Purnell), works for Silco and disrupts Piltover with chaotic acts, including stealing Jayce’s gemstone and triggering a deadly explosion. Enforcer Caitlyn (Katie Leung) frees an imprisoned Vi to find Silco. Jinx's unstable behavior complicates her loyalty, haunted by her past. Meanwhile, Jayce’s rise to the council and Viktor’s experiments with Hexcore strain their partnership as Viktor seeks a cure for his illness.
Vi and Caitlyn face numerous obstacles, including battles with Silco’s enforcers and betrayal by Marcus (Remy Hii), who secretly serves Silco. Ekko (Reed Shannon), a childhood friend leading the Firelights gang against Silco, briefly clashes with Jinx. A fight with explosive consequences deepens Jinx’s instability. Heimerdinger and Ekko begin collaborating after Jinx’s attacks. Viktor, desperate for a cure, causes the accidental death of his assistant, fueling guilt.
Jayce attempts peace talks, offering Zaun’s independence in exchange for Jinx. Jinx, influenced by conflicting loyalties, captures Vi, Caitlyn, and Silco. In a tense showdown, Jinx kills Silco after a confrontation but fully embraces her new identity. She fires a rocket at Piltover’s council as they vote for Zaun's independence, leaving the city’s fate uncertain.
