Phototoxicity in current imaging techniques leads to the observation of perturbed cell and organelles dynamics. Stem cells cultures, primary cell cultures and organelles like mitochondria are highly sensitive to experiment induced stresses like phototoxicity in fluorescence microscopy.
This footage features Primary Human Keratinocytes kindly provided by PromoCell GmbH. In keratinocytes, mitochondria play a role in differentiation[1], [2] and their dysfunctions are involved in many skin diseases[3].
The 3D Cell Explorer allows for the observation of unperturbed mitochondrial dynamics, thanks to its non-invasive technology. By injecting the sample ~100 times less energy (~0.2 nW/µm2) than light sheet microscopes (~1nW/µm2), one can observe details of fusions, fissions and mitochondrial network remodelling.
