Services on this route usually operate every half hour with services having the same stopping pattern. A limited number of services operate direct from Cambuslang to Motherwell skipping Uddingston and Bellshill. A very limited service operates via Holytown between Bellshill and Wishaw skipping Motherwell and Shieldmuir. Additional services operate every hour between Glasgow Central and Bellshill as part of the Glasgow Central to Edinburgh via Shotts service. Cambuslang has additional services to Glasgow Central low level on the Argyle Line from Whifflet to Dalmuir. Motherwell has a slower half hourly Glasgow Argyle Line service via Hamilton Central. There are a few additional trains a day as far as Carluke which operate to Edinburgh via Carstairs. There are connections at Glasgow Central and Motherwell for Avanti West Coast, Cross Country, LNER, Caledonian Sleeper and TransPennine Express services and the Glasgow Subway St Enoch station is a short walk from Glasgow Central station.

The rolling stock on this route is mainly class 385 Hitachi trains however older EMU class 318 and class 320 still operate some services and services are usually formed of 3 or 6 carriages.

This route is fully electrified using overhead AC electrification. This route is mainly at least double track with single track sections between Motherwell and Wishaw and between Carluke and Lanark. Newton station is on a loop and is not served by Lanark trains instead having services via Cathcart and Argyle Line services.
