Angeliki Malliri and colleagues show that for mammalian epithelial cells to divide in the correct orientation and form normal luminal structures, Dlg1 must localise to the cell cortex, which is dependent on binding to CASK. In the first clip, an epithelial cyst grown on Matrigel shows normally oriented cell divisions. In the second clip, CASK is knocked out, and in the third, Dlg1 is knocked out, and abnormally oriented cell divisions are observed.
