Antibodies can be engineered to recognize a wide range of biological targets and have applications in basic research, diagnostics, and therapeutics. The limited diversity of the amino acids found within conventional antibodies constrains antibody properties such as potency, duration of action, and range of targetable epitopes. This presentation will describe our integration of noncanonical amino acids into yeast display to construct chemically expanded antibodies. We have used this platform to discover covalent antibodies and “hybrid” protein-small molecule conjugates with properties that would be nearly possible to access with conventional antibodies.
