The DNS method for estimating the concentration of reducing sugars in a sample was originally invented by G. Miller in 1959.
This method tests for the presence of the free carbonyl group in reducing sugars.
One mole of sugar will react with one mole of 3, 5- dinitro salicylic acid.
The absorbance measured using a spectrophotometer is directly proportional to the amount of reducing sugar. Different reducing sugars generally yield different color intensities; thus, it is necessary to calibrate for each sugar.
Oxidation: Aldehyde group to carboxyl group
Reduction: 3, 5-dinitrosalicylic acid to 3-amino, 5-nitrosalicylic acid
