The LMB Mass Spectrometry and Proteomics Facility houses a wide range of mass spectrometers enabling biological proteomics and structural proteomics. Biological proteomics involves the identification of proteins and their interactions and the comparative proteomics of cells and tissues. In structural proteomics purified proteins or their complexes are investigated by hydrogen-deuterium exchange or cross-linking mass spectrometry.
In this video Mark describes a peptide centric approach to structural proteomics. In the first part of the talk, he explains the theory, typical workflow and data analysis involved in hydrogen-deuterium exchange mass spectrometry. How this approach can reveal conformation changes is illustrated with examples of LMB projects including a post-translation modification, mechanism of activity, and the effect of membrane binding. In the second part of the talk, Mark shows how chemical crosslinkers can be used as a tool for structural investigation using an example of the mitochondrial complex I.
