Pthumeru ihyll layer 3 up to 2:14 (pre made) Rotted pthumeru ihyll root 2:15 onwards. The 2nd method is much faster but requires you to have created a rotted foetid ihyll root dungeon at least once before. The only other place you can get yellow backbones is from the one reborn boss fight which i believe gives 3.
For the root locaiton you must first open the boss gate. The dungeon is small so you should find the lever pretty fast. As for advice on the enmies do not let them double team you. They are very hard to take on at the same time because of there long reach. They also deal alot of damage. Using a pebble might help seperate them. The two handed variant is easy to counter, the ones with the shield are a bit more difficult so i recommend just attacking them. If you hit these enemies while in mid attack it will stop there attack the majority of the time.
