After you rescue DD as a pup, the fastest way to have him available as a buddy is to visit Mother Base as often as you can. Whenever you complete a Story Mission or a few side-ops, return to MB and your DD pup will welcome you as you touch down. After a while, he'll stop appearing, but keep returning to Mother Base.
After a bunch of fly-by's, a cutscene will appear where Ocelot will introduce the fully grown DD and explain his abilities.
