A gap junction hemi-channel is a hexamer, and each protomer consists of 4 transmembrane helices with both N and C termini at the cytoplasm. Both of the 2 extracellular loops interact with the same loops of another hexamer on another cell to form a 12meric gap junction channel. The cytoplasmic entry funnel of the channel is positively charged.
