DNA sequencing is a process of reading the nucleotide bases in a DNA molecule. It includes any method or technology that is used to determine the order of the four bases - A,T,G,C in a strand of DNA. Basically, DNA sequencing precisely determines the order of nucleotides in a piece of DNA.
Maxam Gilbert Method of DNA sequencing uses chemical approach to read the nucleotide sequence and that is why it is also known as chemical cleavage method of DNA sequencing. It was developed by Allan Maxam and Walter Gilbert in 1970s.
In Maxam Gilbert method, the ssDNA fragment to be sequenced is labeled with radioactive phosphate at 5' end which is then divided into four aliquots; each of which is treated with a reagent which modifies a specific base. This will result in strands of varying lengths. These strands are separated out with electrophoresis and is read with radioautography.
