Human pluripotent stem cells (hPSCs) are an excellent resource for studies of cell fate specification, disease modeling and drug screening. In order to produce various neural cells from hPSCs, the induction to neural stem cells (NSCs) is the first important step. Conventional methods of NSC derivation involve embryoid body (EB) formation or co-cultures with stromal cell lines, which has several disadvantages such as long duration, difficulty to control the quality of derived NSCs and the contamination of non-neural cells. To address the need for an easy and scalable process for the NSC generation, we have developed PSC Neural Induction Medium which can convert hPSCs into NSCs in one week with 80-90% of efficiency but without the time consuming laborious processes of embryoid body formation and mechanical NSC isolation. Resultant NSCs show relevant Sox1, Sox2 and Nestin markers, minimal pluripotent marker Oct4 expression and can be subsequently differentiated into neurons and glia.
