Mona-Lisa Malarte, PhD Candidate, Center of Alzheimer Research, Karolinska Institutet, Stockholm, Sweden, explains the results from her study using a multi-PET tracer approach to identify distinguishing features that may aid the differential diagnosis and early intervention of 4R tauopathies. The complex neuropathology and common hallmarks in these diseases makes it difficult when differentiating between them in clinical diagnosis. This study utilizes novel tau and astrocytic tracers to investigate the expression and distribution of these hallmarks in corticobasal degeneration (CBD), progressive supranuclear palsy (PSP), and Alzheimer’s disease (AD) brains. The results highlight the complex nature of these different pathologies, with an array of tau fibrils being expressed across different diseases, as well as within brain regions and cell types of the same disease. This emphasizes the ongoing need for more precise tracers capable of distinguishing between different tau fibril isoforms. Ms Malarte touches on the importance of developing tracers that can identify disease hallmarks that precede the more commonly observed amyloid and tau accumulations, which tend to manifest in later stages of disease.
