To do the Melee Attack while riding a bike, first select your Melee Weapon from the options by tapping 'X' on Xbox ('Square' on PlayStation, 'Tab' on PC) until you have a Melee weapon, then if on Xbox hold down the 'A' button and then LB or RB depending which side you attack (PlayStation is hold 'X' and L1/R1, PC is hold 'X' and left/right mouse). This will usually take people out with one hit, especially if you have some momentum, and even works when you have a friend on the back of your bike.
